 You're thinking of the "Reply-To" header.

the "Reply-To" header.

Nope.
The "Reply-To" header contains the email address to which any reply to that email should be sent. One of my current annoyances is that Telus (my email ISP) contracted with Google to provide my email servces and Google refuses to let me set my "Reply-To" to the email I want to use going forward.
The "In-Reply-To" header IS optional (but "should" be used), but its purpose is to reference the "Message-ID" (also optional but "should" be used) of the email to which the email is a... ...reply.
