Volgens de beschrijving in Genesis 2:10-14 lag de Hof van Eden in een gebied waar vier rivieren ontsprongen, waaronder de Eufraat en de Hiddekel (Tigris). Dit wijst op een locatie ergens in het oude Mesopotamië, wat overeenkomt met het huidige Irak en delen van Syrië, Turkije en Iran.
De exacte locatie is onbekend, maar sommige onderzoekers vermoeden dat het gebied ergens lag in het oude Mesopotamië, wat nu ruwweg Irak en de omliggende regio’s omvat. De Tigris en de Eufraat bestaan nog steeds, maar de andere twee rivieren (Pison en Gichon) zijn moeilijk te identificeren.
Sommige geleerden denken dat de Hof van Eden verloren is gegaan door de Zondvloed in de tijd van Noach (Genesis 6-8), waardoor het landschap ingrijpend veranderde.
